- Sivir: Spell Shield (3rd) blocks one spell, not the true damage your Silver Bolts (2nd) keep stacking onto her, and her short range cannot kite you once you commit to a long fight.
- Kalista: Kalista's hops reposition after attacks but do not make targeted basic attacks miss. Keep Silver Bolts (2nd) on Kalista rather than switching targets, use Tumble (1st) to maintain range, and respect Rend's (3rd) burst before the third stack lands.

- Lulu: She solves your weak lane phase: shields and Whimsy (2nd) let you survive to your item spikes, then Wild Growth (Ult) turns your late-game duels into unloseable fights.
- Janna: Her disengage buys the scaling time your kit demands, and her shield plus your Tumble (1st) spacing makes you nearly impossible to run down before you spike.
- Thresh: Lantern saves cover your aggressive Tumble (1st) angles, and his hooks create the isolated 1v1s where Silver Bolts (2nd) win every time.
Early Game
Protect your health while farming at the edge of Vayne's short range. Use Tumble (1st) to dodge the spell that starts the enemy trade, then take a second attack only when it does not pull you into the support's control. Save Condemn (3rd) for a wall stun, diver, or gank rather than spending it as poke. Against bullies, give up a minion before losing enough health to be forced out of lane. Against a weaker short-range duo, Silver Bolts (2nd) can still reward a three-hit trade when your support controls the return damage.
Mid Game
Once items arrive, activate Silver Bolts (2nd) before an extended fight and keep farming until the map gives a safe objective rotation. At six minutes, follow lane priority rather than crossing an unwarded river to contest by habit. Herald is usually the stronger even trade, while dragon is worth committing to when your composition can protect Vayne through repeated fights toward soul. Final Hour (Ult) adds attack damage, raises Night Hunter's (Passive) chase speed to 70, and makes each Tumble (1st) grant one second of invisibility that ends on attacking or casting. Use that brief stealth to change direction, then look for takedowns that extend the ultimate by four seconds up to its original duration.
Late Game
Three items and Final Hour (Ult) make Vayne a strong front-to-back carry, but short range still decides which targets are safe. Stay behind your frontline, attack the closest threat, and use Tumble's (1st) invisibility to break targeting or change angle instead of rolling deeper by habit. Silver Bolts (2nd) handles high-health targets without requiring you to cross the frontline. Hold Condemn (3rd) for the diver or wall stun that preserves your attack uptime, and arrive early enough to Baron or Elder that you are not forced to walk through fog.
Teamfights
Stand behind your peel and attack the closest target, since Silver Bolts (2nd) makes a tank productive damage rather than an obstacle to bypass. Tumble (1st) between attacks only when the new position preserves spacing, and use Final Hour's (Ult) brief invisibility to break targeting before revealing with the next shot. Hold Condemn (3rd) for the diver who reaches you or a reliable wall stun on a priority target. Do not chase through the enemy formation.
